I have an Ethernet run from the FiOS box to the Router. Can I split that before the router so one Ethernet cable to the Router and another to a computer.
I have the box in my basement with an Ethernet run to the first floor living room where the router is and another run from the basement to the second floor home office. Do I need to connect the home office Ethernet to the Router or can I just split from the box?

No. You have to come out of the LAN port of your router. You can’t split off the ONT. You could also possibly use MoCA adapters if the house is wired up with coax.
