I have Fios Gigibit Internet. I just ordered the G3100 router, I have an Ethernet line and coax coming into the room. Should both be connected into the G3100 to set it up?

That depends. If you have TV services, then yes. If not, you don't need the coax coming from the ONT.

Are you using any service other than internet? If only internet and you have been transitioned over to ethernet only then the ethernet will give you internet connectivity.
If you have tv I believe the coaxial has to be connected as well for tv signal and guide. Alas I don't have Verizon's TV or phone service just gigabit service.
